A series of N-glycan reference standards compatible with Waters GlycoWorks™ RapiFluor-MS™ workflows has been developed for direct and robust identification of N-glycans released from immunoglobulins. The standards include eight bi-antennary N-glycans, with isomers differentiated by sialic acid linkage, enabling clear assignment in chromatographic analysis using both fluorescence and mass spectrometry detection. This methodology simplifies peak identification, reduces the need for extended MS analysis, and is adaptable to various HPLC and UHPLC setups, including changes in column chemistry and separation conditions.
Human IgG and monoclonal antibody samples were then processed using a standardized denaturation, reduction, enzymatic glycan release, and fluorescent labeling workflow prior to HILIC-UPLC analysis. The reference standards are used as retention time markers, supporting accurate and reproducible glycan identification in compliance with regulatory requirements for therapeutic antibody characterization.
